Purification of amyloglucosidase from Aspergillus niger

Abstract

In this work, test were carried out to determine the best conditions for the purification of amyloglucosidase from Aspergillus niger NRRL 3122, using a fixed-bed ion exchange chromatography column. The tests were conducted in a purification column containing anion resin DEAE-cellulose. To maximize the purification conditions, were used as variable parameters feeding speed (46 and 8 cm/h) and operating temperature (25 and 10ºC). The best result was observed when a feeding speed of 8 cm/h was employed at a temperature of 10ºC, resulting in a purification factor of 9.9-fold and a recovery of 47.6%.
